Nucleotide exchange and excision technology (NExT) DNA shuffling And Mutational studies on chloramphenicol actetyltransferase I (CATI)

Abstract: Overall stability is the key to application of proteins in technology and therapy. This thesis wants to analyze how structural perturbation by truncation in combination with directed evolution with selection only for activity and subsequent removal of the structural perturbation improves not only activity but also thermal and chemical stability, and even surprisingly positively influences the aggregation behavior.Furthermore this thesis wanted to investigate whether an already thermal and chemical stable enzyme with a two substrate mechanism and finely tuned interaction between subunits during catalysis can be further stabilized without the requirement of structural information or further hyperthermophilic homologs by structural perturbation and compensation strategy
