The function of WINDHOSE genes during Megasporogenesis in Arabidopsis thaliana

Abstract: When preparing sexual reproduction, a group of cells is specified as germline, that generates reproductive cells i.e. sperms or egg cells and pass their genetic materials onto offspring (Weismann, 1893). Distinct from animals, whose germ cells form at the early stage of embryogenesis (Zwaka and Thomson, 2005), plant germlines are specified in anthers and gynoecia located in flowers after vegetative growth (Schmidt et al., 2015). Their cell identities are seperated from somatic mother tissues. The germ cells are designated to undergo meiosis and mitosis and finally produce haploid egg cells and sperms. However, how the transition from somatic to germline cell fate is regulated during MMC formation is mostly unknown. In the model plant Arabidopsis thaliana, two transcription factors, SPOROCYTELESS/NOZZLE(SPL/NZZ) and WUSCHEL (WUS) is expressed during ovule primordia formation, the targets regulated by them were suspected to participate in the process of a megaspore mother cell establishment (Balasubramanian and Schneitz, 2000; Groß-Hardt et al., 2002). The phytohormone auxin was also reported playing a role in the ovule primordia development and embryo sac formation (Pagnussat et al., 2009). In this study, I focused on the function of the plant specific proteins WINDHOSE1 (WIH1) and WIH2, the downstream genes of WUS, that form a complex together with RAB GTPASE HOMOLOG A1F (RABA1F) and RABA1G (Lieber et al., 2011). All of these proteins are required for the polar PIN1 localization and the auxin response in the epidermal cells of the ovule primordium from where they are required for non-cell-autonomous regulation of germline initiation
