The influence of the universally conserved GTPase YchF on translation and bacterial stress responses

Abstract: The two main aims of this study were the investigation of the questions whether the universally conserved P-Loop GTPase YchF is involved in a mechanism that is part of the regulation of the general stress response master regulator RpoS and what role YchF plays during translation. The model organism E. coli was chosen for the investigations. In this study it could be shown that the levels of rpoS mRNA and RpoS protein are prematurely elevated in exponentially growing ychF deletion cells and that the mRNA levels of the two RpoS target genes katE and osmY are prematurely elevated as well. This leads to the conclusion that YchF might be involved in the regulation of RpoS and that the observed premature upregulation of the expression of the catalase KatE in ychF deletion cells might explain the H2O2 resistance phenotype of ychF deletion cells, known from former studies. The investigation of the role of YchF in translation showed that YchF is involved in the regulation leaderless mRNA translation and in the regulation of the translation interfering type II toxin/antitoxin system MazE/F
