Role of BAX complexes in apoptosis signaling
Abstract: OMM permeabilization by active BAX and BAK results in release of proteins, such as cytochrome c, from the mitochondrial intermembrane space to the cytosol. Cytochrome c mediates apoptosome formation and, thus, triggers activation of the caspase cascade by activation of the initiator caspase-9. On the one hand, small active BAX complexes were reported to be sufficient for efficient OMM permeabilization and protein release. On the other hand, active BAX was shown to form large oligomeric structures, which protrude into the cytosol. In this study, organization of endogenous BAX into either small complexes and large membrane-associated structures was shown. These results demonstrate, that small mitochondrial BAX complexes are sufficient for OMM permeabiliation and cytochrome c release. OMM permeabilization alone did not induce full caspase activation in absence of large BAX scaffolds. Predominant formation of large BAX scaffolds results in impaired OMM permeabilization, but activation of caspase-9 through direct interactions. Therefore, the apoptosome and BAX scaffolds may present redundant activation platforms for caspase-9, ensuring full caspase activity
