A Chromogenic Antithrombin Assay with a Recently Introduced Test Kit Optimization and Titration of Reagents
Since Blomback et al presented the first antithrombin method utilizing a chromogenic substrate (1), there has been a considerable development in this field. In order to evaluate the possibility to recommend a well standardized and reliable procedure, we have tested all the parameters in the assay originally developed by Odegård et al. By using these data the different stages, viz 1) thrombin inhibition by blood plasma and 2) determination of thrombin not consumed, have been optimized. The method proposed can, by using the given data, be modified to suite most kinds of equipment. The reproducibility and stability of the different reagents and standards have been studied and found to be good. The amount of thrombin used in the assay has been titrated by using different methods and the molar amount of active antithrombin in normal human plasma has been calculated. 1) Blombäck M. et al, Thromb.
