Fungal protein analysis by electrospray mass spectrometry : = Pilz-Protein-Analyse durch Elektrospray-Massenspektrometrie

Zusammenfassung: The goal of the work was to develop an analytical protocol based on mass spectrometry aimed at the identification of the protein content of complex biological samples, speeding up biological and medical studies, and thus the diagnoses. More specifically, a method was developed that -without the need of separation techniques or elaborade and time-consuming sample preparation-allows to analyze, identify and possibly quantify as many proteins present in a complex mixture as possible in a continuos way using only the analysis by electrospray ionization mass spectrometry. To apply and test our protocol, two different types of cultures were used: 1. Samples from bone marrow culture serum obtained from the University Hospital of Basel (Switzerland). With these samples, the focus was on the determination of the protein contents and their different peptide interactions, in an approach of combining different methods of proteomic studies on a single protocol. 2. In a second step, a number of cultures were prepared from three different species of fungi (Sacheromyces cerevisiae, Geotrichum candidum and Yarrowia lipolytica). The challenge in these samples was the number and variety of proteins in fungi. In this second set, we used labeling techniques, in addition to the techniques applied in the first group. As a result, the analytical protocol proved to be quite effective for a quick assessment of the protein content of serum mixtures, and in addition it also made possible the identification of the majority of proteins by correlation of the mass spectrometry data with protein data bases
Zusammenfassung: Das Ziel der Arbeit war es, ein analytisches, massenspektrometrisches Protokoll für die Identifizierung des Proteingehalts von komplexen biologischen Proben zu entwickeln, um biologische und medizinische Studien, und damit die Diagnostik im allgemeinen zu beschleunigen. Genauer gesagt, wurde ein Verfahren entwickelt -ohne die Notwendigkeit von Trennverfahren oder aufwendiger und zeitintensiver Probenvorbereitung-, um Proteine in einem komplexen Gemisch in einer kontinuierlichen Weise unter Verwendung von Elektrospray-Ionisation-Massenspektrometrie zu identifizieren und gegebenenfalls zu qualifizieren.
