Photosynthesis - Purple Bacteria: Van Niel's Isolation Technique (Anaerobic Pure Culture)

By serial dilution of a drop of raw culture of mixed purple bacteria various strains can be isolated. With progressively decreasing concentration, the bacteria grow anaerobically in a tube of agar medium (with sulphide and bicarbonate). A light cabinet serves as an incubator. A pure culture is obtained by subjecting a droplet of this to a second serial dilution.
