Die Charakterisierung der funktionellen Interaktion von Neph1 und Scribble im Podozyten

Zusammenfassung: Scribble konnte als neues podozytäres Polaritätsprotein identifiziert werden. Es kolokalisiert im sich differenzierenden Glomerulum mit Neph1. Darüber hinaus konnte gezeigt werden, dass Scribble mit seiner dritten oder vierten PDZ-Domäne eine Interaktion mit dem PDZ-Domänen-Bindemotif von Neph1 eingeht. Diese Interaktion kann durch eine Threonin-Phosphorylierung von Neph1 reguliert werden. Scribble lokalisiert während der glomerulären Entwicklung zunächst basal der Zell-Zellkontakte an denen Par3 lokalisiert, wandert entlang der lateralen Zellmembran nach basal, und kolokalisiert im Glomerulum partiell mit Par3. Scribble kolokalisiert zu keiner Zeit der glomerulären Entwicklung mit dem apiklen Marker Podocalyxin
Zusammenfassung: Scribble was identified as a new protein of podocyte polarity. It colocalizes in the developing glomerular with neph1. Besides this it was shown that scribble interacts with its third or fourth PDZ-domain with the PDZ-binding-motif of neph1. This interaction can be regulated through threonine phosphorylation of neph1. During glomerular development scribble localizes initially basal the cell-cell-contacts and with it basal of par3, later during glomerular development it localizes along the lateral cell membrane and finally at the basal cell membrane. It colocalizes partially with par3. Scribble colocalizes at no point of the glomerular development with the apical protein podocalyxin
