Drei-Kammer-Shuttle PCR für polymerbasierte, druckgetriebene Lab-on-a-Chip Systeme

Zusammenfassung: In this paper two central fields of application are presented. On the one hand a three chamber shuttle based PCR concept is shown and characterized, which offers the possibility for an implementation of multiplex PCR Assays on lab on a chip (LoC) platforms in less than 30.minutes. On the other hand novel laser based production technologies for assembling complex microfluidic platforms, currently under development at Robert Bosch GmbH, are presented.First area of application is located in the field of the development of platform technology, based on polymer materials polycarbonate (PC) and polyurethane (TPU). The domain for LoCs is located in industrial products, so requirements like unit costs, costs of production, time of production referred to mass production have to be fulfilled. Main task is to realize complex, pneumatically forced microfluidic systems in harsh environment. Multifactorial requirements for the laser based assembling process are defined by the PCR concept, which is presented in this work as well. The challenge is to ensure temperature- and pressure stability of the laser welded area (<500.μm diameter) by fulfilling a process time less one hour.
