Effect of Nicotine on Occlusive Arterial Thrombosis in Rats
The effect of nicotine on occlusive arterial thrombosis in Wistar rats from either sex was studied. Arterial thrombi were induced by insertion of polyethylene loop in the abdominal aorta. Obstruction time (OT), dry thrombus weight (TW) and mortality rate (MR) served as criteria for assessment of thrombosis. Nicotine dihydrochloride (0.3, 3 and 6 mg/kg, s.c, dailyfor four weeks) deteriorated in a dose-dependent manner the process of thrombosis in both male and female rats. In comparison with control animals nicotine-treated male as well as female rats showed statistically significant decrease of OT and increase of TW and MR. Nicotine dihydrochloride abolished the previously observed sex difference in arterial thrombosis in rats. These data suggest that nicotine, one of the chief constituents of tobacco smoke, may be considered as an important thrombogenic factor in the development of occlusive arterial thrombosis in rats.
